effectiveness of conventional cancer therapies has driven the development of strategies that harness the potential of the immune system to fight cancer.<\/strong><\/p><p>Immunotherapy is based on various approaches that mobilize immune cells to destroy malignant cells. One such approach is adoptive cell therapy (ACT), which involves administering tumor-specific T lymphocytes capable of killing cancer cells.<br> <\/p><p>Advances in cell-engineering technologies have enabled the generation of modified immune cells in vitro, characterized by enhanced cytotoxic activity. To date, six ACT therapies based on genetically engineered patient-derived T lymphocytes (so-called CAR-T cells\u2014chimeric antigen receptor T cells) have been approved for clinical use in selected blood cancers. Further improvements to ACT include identifying more effective effector cells that do not require autologous administration. One promising candidate group is \u03b3\u03b4 T lymphocytes.<br>   <\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t<div class=\"elementor-element elementor-element-29ea575 e-con-full cut-off-wrapper-90 e-flex e-con e-child\" data-id=\"29ea575\" data-element_type=\"container\" data-e-type=\"container\" data-settings=\"{&quot;background_background&quot;:&quot;classic&quot;}\">\n\t\t\t\t<\/div>\n\t\t<div class=\"elementor-element elementor-element-6cebe7f e-con-full e-flex e-con e-child\" data-id=\"6cebe7f\" data-element_type=\"container\" data-e-type=\"container\" data-settings=\"{&quot;background_background&quot;:&quot;classic&quot;}\">\n\t\t\t\t<div class=\"elementor-element elementor-element-438d316 elementor-widget elementor-widget-text-editor\" data-id=\"438d316\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p><strong data-start=\"1136\" data-end=\"1269\">\u03b3\u03b4 T cells constitute 1\u201310% of circulating T lymphocytes and are considered enigmatic cells that bridge innate and adaptive immune responses.<\/strong><\/p><p>This feature makes them more suitable for immunotherapeutic applications than the dominant \u03b1\u03b2 T lymphocytes, which require antigen processing and presentation by major histocompatibility complex (MHC) molecules. \u03b3\u03b4 T-cell receptors (\u03b3\u03b4 TCRs) can directly recognize molecular patterns of cellular pathology (so-called stress antigens) without the need for MHC involvement.<br> <\/p><p>Importantly, the lack of MHC restriction allows allogeneic use of \u03b3\u03b4 T cells in situations where expansion of autologous T cells is difficult or impossible. Upon activation, \u03b3\u03b4 T cells secrete interferon gamma (IFN-\u03b3) and tumor necrosis factor alpha (TNF-\u03b1)\u2014cytokines well known for their cytotoxic properties. It has been shown that the dominant \u03b3\u03b4 T-cell subpopulation in blood, containing the V\u03b42 chain in the TCR complex, responds to phosphoantigens, which are non-peptide intermediates of the mevalonate and non-mevalonate pathways of isoprenoid biosynthesis.  <\/p><p>The second major \u03b3\u03b4 T-cell subpopulation, containing the V\u03b41 chain and found mainly in the skin and mucosal tissues, recognizes molecules present on stressed and cancerous cells, such as MICA\/B and UL16-binding proteins.
