The research has received <strong data-start=\"335\" data-end=\"354\">PLN 3.5 million<\/strong> in funding from the <strong data-start=\"375\" data-end=\"393\">Weave-U NISONO<\/strong> program.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t<div class=\"elementor-element elementor-element-bd21cd1 e-con-full e-flex e-con e-parent\" data-id=\"bd21cd1\" data-element_type=\"container\" data-e-type=\"container\">\n\t\t<div class=\"elementor-element elementor-element-9f67367 e-grid e-con-boxed e-con e-child\" data-id=\"9f67367\" data-element_type=\"container\" data-e-type=\"container\">\n\t\t\t\t\t<div class=\"e-con-inner\">\n\t\t\t\t<div class=\"elementor-element elementor-element-f69b596 elementor-widget__width-initial elementor-widget elementor-widget-text-editor\" data-id=\"f69b596\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p data-start=\"404\" data-end=\"725\">According to the <strong data-start=\"421\" data-end=\"450\">World Health Organization<\/strong>, by the end of 2023 approximately <strong data-start=\"485\" data-end=\"508\">39.9 million people<\/strong> worldwide were living with HIV, including about <strong data-start=\"557\" data-end=\"591\">1.4 million children aged 0\u201314<\/strong>. These figures raise an important question: why, despite technological progress and advances in medicine, does HIV remain undefeated?<\/p><hr data-start=\"727\" data-end=\"730\" \/><h3 data-start=\"732\" data-end=\"758\">An Enemy Lying Dormant<\/h3><p data-start=\"759\" data-end=\"1146\">After entering a cell, the viral genetic material (RNA) undergoes reverse transcription into viral DNA, enters the cell nucleus, and integrates into the host genome. The virus becomes indistinguishable from our own DNA and can replicate together with host cells. At this stage, some viruses enter a silent infection mode known as latency, making the virus invisible to the immune system.<\/p><p data-start=\"1148\" data-end=\"1474\">\u2013 <em data-start=\"1150\" data-end=\"1358\">What is most dangerous about this virus and what makes it so difficult to eliminate is the reawakening of proviruses from latency\u2014the so-called viral rebound\u2014after discontinuation of antiretroviral therapy,<\/em> explains <strong data-start=\"1368\" data-end=\"1391\">Dr. Heng-Chang Chen<\/strong>, leader of the <strong data-start=\"1407\" data-end=\"1447\">Quantitative Virology Research Group<\/strong> at <strong data-start=\"1451\" data-end=\"1473\">\u0141ukasiewicz \u2013 PORT<\/strong>.<\/p><hr data-start=\"1476\" data-end=\"1479\" \/><h3 data-start=\"1481\" data-end=\"1506\">HIV in the Crosshairs<\/h3><p data-start=\"1507\" data-end=\"1781\"><strong data-start=\"1507\" data-end=\"1519\">Dr. Chen<\/strong> gained experience as a molecular virologist at, among others, <strong data-start=\"1582\" data-end=\"1615\">Humboldt University of Berlin<\/strong>, <strong data-start=\"1617\" data-end=\"1694\">Dr. Filion\u2019s laboratory at the Centre for Genomic Regulation in Barcelona<\/strong>, and <strong data-start=\"1700\" data-end=\"1780\">Dr. Benkirane\u2019s laboratory at the Institute of Human Genetics in Montpellier<\/strong>.<\/p><p data-start=\"1783\" data-end=\"2168\">Scientists searching for effective anti-HIV therapies have managed to slow disease progression and extend patients\u2019 lives. However, these treatments are costly, not accessible to everyone, and require lifelong antiretroviral drug use. Although the virus has not yet been completely eliminated from infected individuals, there is hope\u2014<strong data-start=\"2117\" data-end=\"2129\">Dr. Chen<\/strong> is working on an alternative solution.<\/p><hr data-start=\"2170\" data-end=\"2173\" \/><h3 data-start=\"2175\" data-end=\"2243\">Functional HIV Therapies Combined with Genomic Informatics\u2014Soon?<\/h3><p data-start=\"2244\" data-end=\"2656\">Currently, at least five different antiretroviral strategies are used in the fight against HIV. One of them, the <strong data-start=\"2357\" data-end=\"2377\">\u201cshock and kill\u201d<\/strong> strategy, aims to first reactivate latent HIV and then eliminate it using antiretroviral drugs or the host immune system. Unfortunately, this approach has not been successful to date, prompting scientists and clinicians to explore alternative strategies such as functional cure.<\/p><p data-start=\"2658\" data-end=\"3403\">\u2013 <em data-start=\"2660\" data-end=\"2789\">The \u201cblock and lock\u201d strategy aims to block proviral transcription and permanently lock the reservoir in a deeply latent state,<\/em> explains <strong data-start=\"2799\" data-end=\"2811\">Dr. Chen<\/strong>. <em data-start=\"2813\" data-end=\"3123\">Interestingly, we have observed that different antiretroviral drugs target different subgroups of proviruses. Since establishing my laboratory at \u0141ukasiewicz \u2013 PORT, one of my main research areas has been investigating the impact of the host genome on shaping the microenvironment of the latent HIV reservoir<\/em> (Chen, <em data-start=\"3131\" data-end=\"3141\">Vaccines<\/em> 2023; Wi\u0119cek &amp; Chen, <em data-start=\"3163\" data-end=\"3173\">iScience<\/em> 2023; Wi\u015bniewski et al., <em data-start=\"3199\" data-end=\"3209\">iScience<\/em> 2024). <em data-start=\"3217\" data-end=\"3403\">If we can understand these mechanistic interactions, we may be able to use knowledge about chromatin\u2014such as chromatin silencing\u2014to permanently block HIV transcription in deep latency.<\/em><\/p><p data-start=\"3405\" data-end=\"3648\">The innovation in <strong data-start=\"3423\" data-end=\"3437\">Dr. Chen\u2019s<\/strong> research lies primarily in approaching HIV at a therapeutic and functional genomic level. Locking HIV in a latent state would be a long-awaited breakthrough, freeing patients from the need for daily medication.<\/p><hr data-start=\"3650\" data-end=\"3653\" \/><h3 data-start=\"3655\" data-end=\"3671\">A Clear Goal<\/h3><p data-start=\"3672\" data-end=\"4143\">\u2013 <em data-start=\"3674\" data-end=\"3868\">If the entire genome could be viewed as part of the host\u2019s vast immunological machinery, understanding the functional properties of the genome would be crucial for overcoming viral infections,<\/em> explains <strong data-start=\"3878\" data-end=\"3890\">Dr. Chen<\/strong>. The <strong data-start=\"3896\" data-end=\"3936\">Quantitative Virology Research Group<\/strong> hopes that understanding the role of the host genome in HIV latency will lead to better antiretroviral therapeutic strategies and, ultimately, enable the complete elimination of HIV infection in the future.<\/p><p data-start=\"4145\" data-end=\"4471\">\u2013 <em data-start=\"4147\" data-end=\"4225\">In a scientist\u2019s work, having a clearly defined goal is extremely important,<\/em> emphasizes <strong data-start=\"4237\" data-end=\"4249\">Dr. Chen<\/strong>, for whom the priority\u2014both in his own work and that of the entire team\u2014is to improve the quality of life of people living with HIV by ensuring better disease control and access to more effective and affordable therapies.<\/p><hr data-start=\"4473\" data-end=\"4476\" \/><h3 data-start=\"4478\" data-end=\"4492\">References<\/h3><p data-start=\"4493\" data-end=\"5036\">Chen, H.-C.
