for performing everyday life activities such as eating, speaking, and breathing. The integrity of the sarcolemma (muscle membrane) is maintained by the <strong data-start=\"562\" data-end=\"614\">dystrophin-associated glycoprotein complex (DGC)<\/strong>. The functioning of the DGC is impaired in many muscular dystrophies. For example, the absence of <strong data-start=\"713\" data-end=\"727\">dystrophin<\/strong>, caused by mutations in its encoding gene, is the primary cause of <strong data-start=\"795\" data-end=\"832\">Duchenne muscular dystrophy (DMD)<\/strong>. Currently, there is no cure for DMD, and treatment focuses only on alleviating symptoms. Numerous clinical trials on DMD are ongoing, most of which concentrate on restoring dystrophin production in muscles.<\/p><p data-start=\"1044\" data-end=\"1493\">Importantly, the absence of dystrophin leads to the <strong data-start=\"1096\" data-end=\"1136\">degradation of \u03b2-dystroglycan (\u03b2-DG)<\/strong>, a protein that links the extracellular matrix to the actin cytoskeleton. This occurs because, in the absence of dystrophin, the <strong data-start=\"1266\" data-end=\"1280\">PPXY motif<\/strong> on \u03b2-DG becomes exposed and undergoes phosphorylation, resulting in \u03b2-DG degradation. Despite decades of research on DMD, little is known about the proteins that regulate \u03b2-DG stabilization on the cell surface.<\/p><p data-start=\"1495\" data-end=\"1902\">Our research aims to <strong data-start=\"1516\" data-end=\"1583\">identify new proteins that interact with the PPXY motif of \u03b2-DG<\/strong> and stabilize \u03b2-DG by preventing its phosphorylation. Additionally, we will investigate the <strong data-start=\"1676\" data-end=\"1714\">intracellular trafficking pathways<\/strong> of \u03b2-DG during its degradation.
