However, the applicant demonstrated\u2014using <strong data-start=\"612\" data-end=\"636\">cutaneous \u03b3\u03b4 T cells<\/strong>\u2014that autoreactivity can actually be <strong data-start=\"673\" data-end=\"687\">beneficial<\/strong> and play a <strong data-start=\"699\" data-end=\"716\">critical role<\/strong> in maintaining <strong data-start=\"732\" data-end=\"754\">tissue homeostasis<\/strong> and <strong data-start=\"759\" data-end=\"787\">regulating wound healing<\/strong> (Chodaczek et al., <em data-start=\"807\" data-end=\"826\">Nature Immunology<\/em>, 2012).<\/p><p data-start=\"838\" data-end=\"1324\">Through an innovative combination of <strong data-start=\"875\" data-end=\"922\">static and intravital microscopy techniques<\/strong>, it was shown that \u03b3\u03b4 T lymphocytes in the <strong data-start=\"966\" data-end=\"988\">healthy mouse skin<\/strong> are <strong data-start=\"993\" data-end=\"1021\">constitutively activated<\/strong> due to <strong data-start=\"1029\" data-end=\"1090\">continuous stimulation of the \u03b3\u03b4 T-cell receptor (TCR \u03b3\u03b4)<\/strong> within <strong data-start=\"1098\" data-end=\"1124\">immunological synapses<\/strong>. Contrary to the prevailing view that TCR \u03b3\u03b4 recognizes <strong data-start=\"1181\" data-end=\"1218\">undefined stress-induced antigens<\/strong>, these findings suggest the presence of a <strong data-start=\"1261\" data-end=\"1282\">ligand for TCR \u03b3\u03b4<\/strong> that exists even in <strong data-start=\"1303\" data-end=\"1321\">healthy tissue<\/strong>.<\/p><p data-start=\"1326\" data-end=\"1856\">In controlling wound healing, cutaneous \u03b3\u03b4 T cells utilize not only <strong data-start=\"1394\" data-end=\"1401\">TCR<\/strong> but also <strong data-start=\"1411\" data-end=\"1420\">NKG2D<\/strong>, a receptor characteristic of <strong data-start=\"1451\" data-end=\"1463\">NK cells<\/strong>, which recognizes <strong data-start=\"1482\" data-end=\"1519\">proteins induced by tissue injury<\/strong>. However, <strong data-start=\"1530\" data-end=\"1550\">in vitro studies<\/strong> have not definitively established which signaling pathway\u2014TCR or NKG2D\u2014plays a greater role in the <strong data-start=\"1650\" data-end=\"1672\">effector functions<\/strong> of \u03b3\u03b4 T cells and their ability to <strong data-start=\"1708\" data-end=\"1734\">stimulate wound repair<\/strong>. The analysis is complicated by the fact that the <strong data-start=\"1785\" data-end=\"1853\">natural ligand for the skin TCR \u03b3\u03b4 receptor remains unidentified<\/strong>.<\/p><p data-start=\"1858\" data-end=\"2251\">Given the <strong data-start=\"1868\" data-end=\"1892\">unexpected discovery<\/strong> of <strong data-start=\"1896\" data-end=\"1931\">constitutive \u03b3\u03b4 T-cell synapses<\/strong>, it is still unclear whether <strong data-start=\"1961\" data-end=\"1982\">in vitro findings<\/strong> accurately reflect the <strong data-start=\"2006\" data-end=\"2027\">in vivo situation<\/strong>. The proposed project was designed to address these unresolved questions and aims to <strong data-start=\"2113\" data-end=\"2158\">elucidate the role of autoreactive TCR \u03b3\u03b4<\/strong> and the significance of <strong data-start=\"2183\" data-end=\"2207\">cutaneous \u03b3\u03b4 T cells<\/strong> in maintaining <strong data-start=\"2223\" data-end=\"2248\">epidermal homeostasis<\/strong>.<\/p><p data-start=\"2253\" data-end=\"2739\">The central <strong data-start=\"2265\" data-end=\"2288\">research hypothesis<\/strong> assumes that <strong data-start=\"2302\" data-end=\"2327\">constitutive synapses<\/strong> serve as <strong data-start=\"2337\" data-end=\"2398\">sites of intracellular or extracellular material transfer<\/strong>, with such material\u2014stored in <strong data-start=\"2429\" data-end=\"2453\">cytoplasmic granules<\/strong>\u2014either secreted toward <strong data-start=\"2477\" data-end=\"2494\">keratinocytes<\/strong> or internalized by the lymphocyte. It is further hypothesized that the <strong data-start=\"2566\" data-end=\"2605\">dynamics of intracellular transport<\/strong> differ between <strong data-start=\"2621\" data-end=\"2652\">healthy and stressed tissue<\/strong>, depending on the <strong data-start=\"2671\" data-end=\"2692\">activation signal<\/strong> delivered by either <strong data-start=\"2713\" data-end=\"2723\">TCR \u03b3\u03b4<\/strong> or <strong data-start=\"2727\" data-end=\"2736\">NKG2D<\/strong>.<\/p><p data-start=\"2741\" data-end=\"3167\" data-is-last-node=\"\" data-is-only-node=\"\">The research tasks described in this project are designed to test these hypotheses. Previous studies revealed an <strong data-start=\"2854\" data-end=\"2909\">accumulation of GM1 ganglioside-containing vesicles<\/strong> within the synapses. Since <strong data-start=\"2937\" data-end=\"2953\">gangliosides<\/strong> are known to <strong data-start=\"2967\" data-end=\"3006\">induce keratinocyte differentiation<\/strong>, it is hypothesized that <strong data-start=\"3032\" data-end=\"3094\">\u03b3\u03b4 T cells release these vesicles to promote wound healing<\/strong>.
