data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p data-start=\"408\" data-end=\"764\">The ability of the human body to move relies on the <strong data-start=\"460\" data-end=\"516\">cooperation between the nervous and muscular systems<\/strong>. A signal generated in the brain is transmitted through <strong data-start=\"573\" data-end=\"590\">motor neurons<\/strong> to <strong data-start=\"594\" data-end=\"611\">muscle fibers<\/strong>, triggering muscle contraction. This transmission is made possible by a <strong data-start=\"684\" data-end=\"715\">specialized type of synapse<\/strong> known as the <strong data-start=\"729\" data-end=\"761\">neuromuscular junction (NMJ)<\/strong>.<\/p><p data-start=\"766\" data-end=\"1402\">In this synapse, the neurotransmitter <strong data-start=\"804\" data-end=\"821\">acetylcholine<\/strong> is released from synaptic vesicles at the terminal of the motor neuron\u2019s axon. It then diffuses across the synaptic cleft and binds to <strong data-start=\"957\" data-end=\"992\">acetylcholine receptors (AChRs)<\/strong> located on the <strong data-start=\"1008\" data-end=\"1033\">muscle fiber membrane<\/strong>. These receptors are <strong data-start=\"1055\" data-end=\"1075\">highly clustered<\/strong>, and their organization depends on numerous <strong data-start=\"1120\" data-end=\"1144\">scaffolding proteins<\/strong> that interact with the <strong data-start=\"1168\" data-end=\"1193\">cellular cytoskeleton<\/strong>, particularly with <strong data-start=\"1213\" data-end=\"1232\">actin filaments<\/strong>. The actin cytoskeleton undergoes dynamic remodeling in response to internal and external signals. One of the proteins involved in this remodeling process is <strong data-start=\"1391\" data-end=\"1399\">Cap2<\/strong>.<\/p><p data-start=\"1404\" data-end=\"2354\">Previous studies have shown that <strong data-start=\"1437\" data-end=\"1445\">Cap2<\/strong> is essential for the <strong data-start=\"1467\" data-end=\"1520\">development and proper function of cardiac muscle<\/strong>, and its absence causes severe <strong data-start=\"1552\" data-end=\"1577\">cardiac abnormalities<\/strong> in both mice and humans. In skeletal muscles, <strong data-start=\"1624\" data-end=\"1644\">Cap2 dysfunction<\/strong> is associated with <strong data-start=\"1664\" data-end=\"1683\">muscle weakness<\/strong> and the formation of abnormal <strong data-start=\"1714\" data-end=\"1736\">protein aggregates<\/strong> known as <strong data-start=\"1746\" data-end=\"1763\">nemaline rods<\/strong>, which impair muscle function. Our collaborative research with a German group has demonstrated that <strong data-start=\"1864\" data-end=\"1896\">Cap2 knockout (Cap2 KO) mice<\/strong> develop <strong data-start=\"1905\" data-end=\"1920\">ring fibers<\/strong> in fast-twitch muscles. These are <strong data-start=\"1955\" data-end=\"1991\">myofibrils oriented transversely<\/strong> relative to other fibers in the muscle, leading to <strong data-start=\"2043\" data-end=\"2075\">defective muscle contraction<\/strong>. Interestingly, similar ring fibers have been observed in humans with <strong data-start=\"2146\" data-end=\"2178\">myopathies of unknown origin<\/strong>. These findings indicate that <strong data-start=\"2209\" data-end=\"2268\">Cap2 plays a crucial role in skeletal muscle physiology<\/strong>, though its specific <strong data-start=\"2290\" data-end=\"2332\">function at the neuromuscular junction<\/strong> remains unexplored.<\/p><p data-start=\"2356\" data-end=\"2879\">Our preliminary, unpublished results show that <strong data-start=\"2403\" data-end=\"2419\">loss of Cap2<\/strong> in <strong data-start=\"2423\" data-end=\"2439\">Cap2 KO mice<\/strong> leads to <strong data-start=\"2449\" data-end=\"2490\">abnormal motor neuron axon morphology<\/strong>, <strong data-start=\"2492\" data-end=\"2521\">defective AChR clustering<\/strong>, and <strong data-start=\"2527\" data-end=\"2561\">altered synaptic junction size<\/strong>. These findings suggest that <strong data-start=\"2591\" data-end=\"2642\">Cap2 is important for both sides of the synapse<\/strong> \u2014 the <strong data-start=\"2649\" data-end=\"2665\">motor neuron<\/strong> and the <strong data-start=\"2674\" data-end=\"2690\">muscle fiber<\/strong>. To study these roles in more detail, I have generated <strong data-start=\"2746\" data-end=\"2782\">tissue-specific knockout mutants<\/strong>, lacking Cap2 <strong data-start=\"2797\" data-end=\"2833\">only in motor neurons (Cap2 nKO)<\/strong> or <strong data-start=\"2837\" data-end=\"2876\">only in skeletal muscles (Cap2 mKO)<\/strong>.<\/p><p data-start=\"2881\" data-end=\"3284\">Within this project, I will conduct a <strong data-start=\"2919\" data-end=\"2953\">comprehensive characterization<\/strong> of these mutants, focusing on the <strong data-start=\"2988\" data-end=\"3021\">morphology and ultrastructure<\/strong> of neuromuscular junctions using various <strong data-start=\"3063\" data-end=\"3088\">microscopy techniques<\/strong>.
