PORT Polish Center for Technology Development<\/strong><\/p><\/li><\/ul><p data-start=\"533\" data-end=\"651\">The goal of this project is to explore <strong data-start=\"572\" data-end=\"618\">innovative mechanisms for pain suppression<\/strong> while minimizing side effects.<\/p><p data-start=\"653\" data-end=\"991\">In this study, we aim to <strong data-start=\"678\" data-end=\"723\">analyze noradrenergic inhibitory pathways<\/strong> that modulate pain through their <strong data-start=\"757\" data-end=\"827\">direct action on the synaptic terminals of peripheral nerve fibers<\/strong> within the <strong data-start=\"839\" data-end=\"854\">spinal cord<\/strong>. This means that the brain can directly influence the <strong data-start=\"909\" data-end=\"938\">peripheral nervous system<\/strong> through a molecular messenger \u2014 <strong data-start=\"971\" data-end=\"988\">noradrenaline<\/strong>.<\/p><p data-start=\"993\" data-end=\"1437\">We will investigate how <strong data-start=\"1017\" data-end=\"1075\">noradrenaline originating from different brain regions<\/strong> affects <strong data-start=\"1084\" data-end=\"1173\">neurotransmission from the peripheral nervous system to the spinal projection neurons<\/strong>. It is known that nerves possess only one of the nine noradrenaline receptors \u2014 <strong data-start=\"1254\" data-end=\"1264\">Adra2c<\/strong>. In this project, we will <strong data-start=\"1291\" data-end=\"1327\">selectively delete this receptor<\/strong> exclusively in <strong data-start=\"1343\" data-end=\"1370\">peripheral nerve fibers<\/strong> and assess the resulting impact on <strong data-start=\"1406\" data-end=\"1434\">pain signal transmission<\/strong>.<\/p><p data-start=\"1439\" data-end=\"1781\">We will monitor the effects of <strong data-start=\"1470\" data-end=\"1494\">selective modulation<\/strong> of the activity of specific <strong data-start=\"1523\" data-end=\"1548\">noradrenergic centers<\/strong> (using <strong data-start=\"1556\" data-end=\"1572\">optogenetics<\/strong>) through advanced physiological tools such as <strong data-start=\"1619\" data-end=\"1646\">in vivo calcium imaging<\/strong>, <strong data-start=\"1648\" data-end=\"1681\">electrophysiological analysis<\/strong>, and <strong data-start=\"1687\" data-end=\"1728\">computer-based behavioral assessments<\/strong>, conducted <strong data-start=\"1740\" data-end=\"1778\">before and after receptor deletion<\/strong>.<\/p><p data-start=\"1783\" data-end=\"2116\" data-is-last-node=\"\" data-is-only-node=\"\">Ultimately, we hope to <strong data-start=\"1806\" data-end=\"1849\">identify all individual neural pathways<\/strong> through which the <strong data-start=\"1868\" data-end=\"1896\">brain regulates activity<\/strong> in the <strong data-start=\"1904\" data-end=\"1948\">first \u201crelay station\u201d of the spinal cord<\/strong>.
