The major etiological factors contributing to HNSCC development are <strong data-start=\"868\" data-end=\"901\">tobacco use and alcohol abuse<\/strong>, while the incidence is also increasing due to <strong data-start=\"949\" data-end=\"979\">human papillomavirus (HPV)<\/strong> infection.<\/p><p data-start=\"994\" data-end=\"1678\">HNSCCs are anatomically classified based on the <strong data-start=\"1042\" data-end=\"1064\">primary tumor site<\/strong>, which determines treatment strategies. When diagnosed early, the survival rate reaches <strong data-start=\"1153\" data-end=\"1163\">80\u201390%<\/strong>, but approximately <strong data-start=\"1183\" data-end=\"1202\">50% of patients<\/strong> with locally advanced disease experience recurrence, which remains the main cause of treatment failure. Despite improvements in therapies, <strong data-start=\"1342\" data-end=\"1387\">prognosis for HNSCC patients remains poor<\/strong>, and current treatment options are associated with <strong data-start=\"1439\" data-end=\"1462\">severe side effects<\/strong>. Surgical procedures and radiotherapy can impair breathing, swallowing, and speech, while radiotherapy may also cause <strong data-start=\"1581\" data-end=\"1611\">radiation-induced fibrosis<\/strong>, damaging blood vessels that nourish muscles, nerves, and bones.<\/p><p data-start=\"1680\" data-end=\"1950\">At present, <strong data-start=\"1692\" data-end=\"1731\">no clinically applicable biomarkers<\/strong> exist to identify radioresistant tumors, and current <strong data-start=\"1785\" data-end=\"1819\">patient stratification systems<\/strong> for radiotherapy response are insufficient, leading to suboptimal use of healthcare resources and delayed treatment adjustments.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-1fbb009 elementor-widget elementor-widget-heading\" data-id=\"1fbb009\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Project objectives<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-0702144 elementor-widget elementor-widget-text-editor\" data-id=\"0702144\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p data-start=\"1985\" data-end=\"2436\">The project aims to analyze <strong data-start=\"2013\" data-end=\"2087\">digitized hematoxylin and eosin (H&amp;E)-stained histopathological slides<\/strong> using <strong data-start=\"2094\" data-end=\"2148\">digital pathology and artificial intelligence (AI)<\/strong> tools to investigate <strong data-start=\"2170\" data-end=\"2200\">radioresistance mechanisms<\/strong> in HNSCC. Additionally, we plan to explore <strong data-start=\"2244\" data-end=\"2274\">gene\u2013protein relationships<\/strong> using <strong data-start=\"2281\" data-end=\"2324\">advanced molecular pathology techniques<\/strong>, enabling the integration of <strong data-start=\"2354\" data-end=\"2370\">genomic data<\/strong> with <strong data-start=\"2376\" data-end=\"2409\">specific cellular populations<\/strong> within the tumor tissue.<\/p><p data-start=\"2438\" data-end=\"2792\">Through these approaches, we aim to <strong data-start=\"2474\" data-end=\"2516\">identify mechanisms of radioresistance<\/strong> while also elucidating the biological pathways responsible for <strong data-start=\"2580\" data-end=\"2622\">radiation and chemotherapy sensitivity<\/strong> in certain HNSCC subtypes. Both digital and molecular pathology methodologies represent <strong data-start=\"2711\" data-end=\"2750\">innovative and transformative tools<\/strong> in fundamental cancer biology research.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-8261a65 elementor-widget elementor-widget-heading\" data-id=\"8261a65\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Scientific rationale<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-1440156 elementor-widget elementor-widget-text-editor\" data-id=\"1440156\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p data-start=\"2829\" data-end=\"3249\">The <strong data-start=\"2833\" data-end=\"2861\">pathomorphology of HNSCC<\/strong> plays a crucial role in developing resistance to radiotherapy. Premalignant lesions frequently extend to <strong data-start=\"2967\" data-end=\"2987\">surgical margins<\/strong>, and even after tumor removal, they may serve as sources of <strong data-start=\"3048\" data-end=\"3068\">local recurrence<\/strong> or <strong data-start=\"3072\" data-end=\"3094\">new primary tumors<\/strong>. These changes occur within the <strong data-start=\"3127\" data-end=\"3157\">extracellular matrix (ECM)<\/strong> \u2014 a structure providing both <strong data-start=\"3187\" data-end=\"3225\">mechanical and biochemical support<\/strong> to surrounding cells.<\/p><p data-start=\"3251\" data-end=\"3501\">Tumor cells coexist with various <strong data-start=\"3284\" data-end=\"3323\">stromal and immune cell populations<\/strong> (e.g., fibroblasts, endothelial cells, and immune cells) that form part of the <strong data-start=\"3403\" data-end=\"3435\">tumor microenvironment (TME)<\/strong> and modulate cancer progression by remodeling local conditions.<\/p><p data-start=\"3503\" data-end=\"3630\">HNSCCs are characterized by significant <strong data-start=\"3543\" data-end=\"3585\">molecular and phenotypic heterogeneity<\/strong> and harbor numerous <strong data-start=\"3606\" data-end=\"3627\">genetic mutations<\/strong>.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-49c9eb0 elementor-widget elementor-widget-heading\" data-id=\"49c9eb0\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h2 class=\"elementor-heading-title elementor-size-default\">Research hypothesis<\/h2>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-3a5e5cf elementor-widget elementor-widget-text-editor\" data-id=\"3a5e5cf\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p data-start=\"3666\" data-end=\"3955\">We hypothesize that <strong data-start=\"3686\" data-end=\"3710\">not only tumor cells<\/strong> but also <strong data-start=\"3720\" data-end=\"3776\">minor alterations in the seemingly normal epithelium<\/strong>, as well as <strong data-start=\"3789\" data-end=\"3834\">tumor-associated stromal and immune cells<\/strong>, play pivotal roles in regulating <strong data-start=\"3869\" data-end=\"3952\">tumor function, disease progression, and response or resistance to radiotherapy<\/strong>.<\/p><p data-start=\"3957\" data-end=\"4424\" data-is-last-node=\"\" data-is-only-node=\"\">A deeper understanding of the <strong data-start=\"3987\" data-end=\"4013\">tumor microenvironment<\/strong> and its influence on tumor behavior will ultimately enhance our ability to explain and predict <strong data-start=\"4109\" data-end=\"4162\">treatment sensitivity, resistance, and recurrence<\/strong>.
