Similar effects have been observed in <strong data-start=\"1266\" data-end=\"1301\">rodent models of chronic stress<\/strong>, where activation of the <strong data-start=\"1327\" data-end=\"1359\">glucocorticoid receptor (GR)<\/strong> by the stress hormone <strong data-start=\"1382\" data-end=\"1407\">corticosterone (CORT)<\/strong> was implicated. In these models, <strong data-start=\"1441\" data-end=\"1469\">antidepressant treatment<\/strong>\u2014including <strong data-start=\"1480\" data-end=\"1492\">ketamine<\/strong>\u2014has been shown to reverse synaptic deficits in the PFC, suggesting that <strong data-start=\"1565\" data-end=\"1629\">restoration of structural and functional synaptic plasticity<\/strong> is a key mechanism of antidepressant efficacy.<\/p><p data-start=\"1680\" data-end=\"2245\">However, the <strong data-start=\"1693\" data-end=\"1730\">cellular and molecular mechanisms<\/strong> driving <strong data-start=\"1739\" data-end=\"1768\">neural circuit remodeling<\/strong> under stress and depression remain poorly understood. Recent findings by the research team revealed that <strong data-start=\"1874\" data-end=\"1902\">astrocyte-specific genes<\/strong> involved in <strong data-start=\"1915\" data-end=\"1952\">synapse formation and elimination<\/strong> are <strong data-start=\"1957\" data-end=\"1974\">downregulated<\/strong> in the PFC of both MDD patients and chronically stressed rodents. Strikingly, these changes were <strong data-start=\"2072\" data-end=\"2131\">prevented when GR was selectively deleted in astrocytes<\/strong>, indicating that astrocytes may play a <strong data-start=\"2171\" data-end=\"2203\">previously unrecognized role<\/strong> in stress-induced synaptic dysfunction.<\/p><p data-start=\"2247\" data-end=\"2652\">The project aims to <strong data-start=\"2267\" data-end=\"2299\">directly test the hypothesis<\/strong> that <strong data-start=\"2305\" data-end=\"2366\">stress-induced downregulation of astrocyte-specific genes<\/strong> leads to <strong data-start=\"2376\" data-end=\"2399\">synapse loss in MDD<\/strong>. To achieve this, researchers will conduct a <strong data-start=\"2445\" data-end=\"2468\">systematic analysis<\/strong> of the effects of <strong data-start=\"2487\" data-end=\"2524\">astrocyte-specific gene knockdown<\/strong>\u2014targeting those involved in synapse formation or elimination\u2014on <strong data-start=\"2589\" data-end=\"2616\">PFC-dependent behaviors<\/strong>, such as <strong data-start=\"2626\" data-end=\"2649\">social interactions<\/strong>.<\/p><p data-start=\"2654\" data-end=\"3101\">Using <strong data-start=\"2660\" data-end=\"2677\">viral vectors<\/strong> encoding <strong data-start=\"2687\" data-end=\"2707\">shRNA constructs<\/strong>, the team will manipulate astrocyte gene expression and assess behavioral consequences in mice through <strong data-start=\"2811\" data-end=\"2858\">automated behavioral profiling (Social Box)<\/strong>, which monitors approximately 60 spontaneous behaviors. In parallel, astrocytes will be <strong data-start=\"2947\" data-end=\"3017\">genetically perturbed in a cell-type-specific and inducible manner<\/strong> using an innovative <strong data-start=\"3038\" data-end=\"3076\">in utero electroporation technique<\/strong> developed by the team.<\/p><p data-start=\"3103\" data-end=\"3647\">This approach allows for <strong data-start=\"3128\" data-end=\"3155\">high-resolution imaging<\/strong> of <strong data-start=\"3159\" data-end=\"3189\">Thy1-GFP-labeled dendrites<\/strong> in contact with either <strong data-start=\"3213\" data-end=\"3237\">genetically modified<\/strong> or <strong data-start=\"3241\" data-end=\"3265\">wild-type astrocytes<\/strong> within the same animal. Subsequent <strong data-start=\"3301\" data-end=\"3339\">longitudinal two-photon microscopy<\/strong> will track how astrocyte-specific gene silencing affects <strong data-start=\"3397\" data-end=\"3453\">stress- or ketamine-induced dendritic spine turnover<\/strong>. For genes associated with both behavioral and structural changes, <strong data-start=\"3521\" data-end=\"3573\">correlative light and electron microscopy (CLEM)<\/strong> will be used to examine <strong data-start=\"3598\" data-end=\"3634\">astrocyte-synapse ultrastructure<\/strong> in detail.<\/p><p data-start=\"3649\" data-end=\"4107\" data-is-last-node=\"\" data-is-only-node=\"\">By combining <strong data-start=\"3662\" data-end=\"3727\">behavioral, molecular, and high-resolution imaging approaches<\/strong>, this project aims to deliver <strong data-start=\"3758\" data-end=\"3812\">unprecedented insights into the role of astrocytes<\/strong> in <strong data-start=\"3816\" data-end=\"3851\">adult neural circuit remodeling<\/strong>.
