data-end=\"1354\">The aim of the project is to analyze the mechanisms of immune control in the reproductive mucosa, with particular emphasis on the activity of lymphocytes expressing the \u03b3\u03b4 T-cell receptor (\u03b3\u03b4 TCR). The study employs innovative imaging techniques based on high-resolution confocal microscopy and in vivo intravital microscopy. To achieve a comprehensive characterization of immune surveillance\u2014essential for maintaining epithelial homeostasis during infection or injury\u2014the proposed in situ cross-sectional analysis will be complemented with additional advanced ex vivo techniques.<\/p><p data-start=\"1356\" data-end=\"1643\">Among the many cell types involved in mucosal surveillance, \u03b3\u03b4 T cells play a particularly important role. The prevailing view in epithelial immunology is that \u03b3\u03b4 T lymphocytes regulate tissue homeostasis only after recognizing self-antigens that become exposed upon injury or infection.<\/p><p data-start=\"1645\" data-end=\"2004\">However, our previous studies\u2014using a novel combination of intravital confocal microscopy and immunofluorescence techniques (Chodaczek et al., <em data-start=\"1788\" data-end=\"1807\">Nature Immunology<\/em> 2012)\u2014demonstrated that skin-resident \u03b3\u03b4 T cells remain in a continuously activated state due to constitutive phosphorylation of the \u03b3\u03b4 TCR, which physically immobilizes them within the epidermis.<\/p><p data-start=\"2006\" data-end=\"2395\">Contrary to the widely accepted hypothesis, this suggests the continuous expression of specific antigens in healthy tissue. It remains unknown whether similar mechanisms apply to other epithelia also populated by \u03b3\u03b4 T cells.
